IFCB notes KM24 SO-PACE Hawaii to Tahiti, Oct-Nov 2024 Guillaume Bourdin Sample D20241019T233419_IFCB107 to D20241020T051222_IFCB107 taken just after underway was flushed with bleach and rinsed. A lot of detritus in sample, bleach or detritus coming from bleaching the pipes potentially remaining in the line. Rosette S02C01 @5m: - D20241025T162000_IFCB107: a lot of non-living particles, falcon tube likely dirty. - D20241025T172001_IFCB107: Try again with clean tube: good sample, disregard D20241025T162000_IFCB107 Rosette S03C01 @5m: - D20241026T135000_IFCB107: a lot non-living particles, falcon rinsed 3x with DIW + 3x with Niskin water - D20241026T141959_IFCB107: Same problem, maybe coming from the falcon tube or the intake tube. Faulty rotary valve optical sensor, start running IFCB in continuous mode from 2024-10-29 09:00:00 to prevent disconnecting sensor after each sample. D20241029T203818_IFCB107 and D20241029T210210_IFCB107 alignment adjustment during a normal samples, samples still good More particles in UH IFCB, from D20241102T200405_IFCB107 laser focus was changed, one small step to the left (<-500) and binarize threshold was changed to 4 IFCB stopped before D20241108T005200_IFCB107, error when syringe was moving. When IFCB started again with D20241108T005200_IFCB107, the sample had way more detritus, which means there was probably some kind of contamination in the water line when it stopped. Probably still usable, just ignore detritus. Rosette S10C17 @5m: - D20241109T000057_IFCB107: acid washed glass vial and intake tube, but still non-living particles contamination - D20241109T002449_IFCB107: same vial, samples contaminated somewhere Artifact in D20241109T024800_IFCB107 and D20241109T031153_IFCB107: run cleaning just after these samples. A lot of non-living particles found in the following sample due to cleaning Artifact in D20241109T193549_IFCB107, disappeared in D20241109T195941_IFCB107 but run cleaning in case after D20241109T195941_IFCB107. A lot of non-living particles found in the following sample due to cleaning. Again the repeated images appear in: D20241129T163238 D20241129T171757 D20241129T184834 D20241129T191114 did 2x 2ml micro90 run and then three samples fast. images look good from the first sample since then. several good samples and then another two with repeated images: D20241129T225851 D20241129T234409 stopped for cleaning underway after D20241130T070006, resumed at D20241130T093002 Nov 30 all samples from overnight are good except one with repeated image: D20241130T140156 alignment looks good humidity warning at 70%, acquisition stopped, opening IFCB at 23:50 UTC no visible leaks or moisture; replaced desiccant bags and re-canned. when re-started, syringe at exhaust was -1 and the instrument would not run a sample (buttons in software were click-able but did not initiate sampling). tried starting ifcb acquire several times; still not working. did a full power cycle with the switch at the power supply; after that the software started and worked well. first sample after this (D20241201T014155) looks good and shows humidity of 40%. The 10 samples after also have humidity of 40-41. we suspect that the o-ring was not well-seated after the ifcb was opened in port in Tahiti and thus some of the humid lab air was getting into the can. Dec 1 Files from overnight look good re: alignment and images, and humidity is holding at 39%. Only exception is file D20241201T092624, which has the repeated image in it. Large amount of pennate diatoms starting with sample D20241201T155147 Images got out of alignment and focus starting at D20241201T180745 and the four following. Stopping acquisition to run a debubble procedure (modified; document from Guillaume but only the version without opening the can). First sample after debubble (D20241201T204351) looks worse and with big bubbles. Running two more, with a prime intake first. Samples look very bad, terrible alignment, out of focus, and humidity is jumping up. Opening IFCB to inspect. Powering down first to unplug/reposition blue power cable. Upon re-boot the syringe showed -1 at exhaust and no samples would run despite several reboots. Tried cleaning contact at the valve encoder. Eventually stopped trying to wait for info from McLane. Bad samples end at D20241201T213257. Dec 2 Phone call with McLane morning of 12/2 - we isolated the problem to be the communication to the valve sensor and syringe. Unplugged and replugged every related connection. Also tightened the syringe to finger tight; it was loose. IFCB acquire powered up normally with syringe at 0.0 at exhaust. Working well now. Tightening the syringe should also address the humidity in the can issue. Good samples start at D20241202T174741 - many small flagellates, also pennate diatoms - noticed at 06:50 UTC (12/3) that the alignment is shifted off. stopping acq. after current sample to run the modified (in-can) debubble procedure). Starting regular acquisition at 0741 UTC (12/3) Dec 3 Alignment slight off: D20241203T084844, D20241203T091124, D20241203T093404 worse off starting at D20241203T095645 until D20241203T182114 Then a couple slightly off, then looks good starting at D20241203T192916 - stopped for underway cleaning after D20241204T064926; resumed at D20241204T081454 Dec 4 Alignment and humidity are good. Images looked slightly out of focus; adjusted a few times, stopping at -300 - looks better now. Dec 5 Stopped for underway cleaning at 06:13 UTC (12/6) Dec 7 Stopped for underway cleaning at 06:22 UTC (12/8). File from 06:13 is of fresh tap water (forgot to stop IFCB first). BAD FILE: D20241208T061334. Deleted from IFCB computer, Cetautomatix, and hard disk, but still shows in the time series view of the dashboard (but images do not show). Started IFCB again after cleaning at 07:32 UTC 12/8 Dec 9 19:06 UTC switched from 1 ml to 3 ml prime before intake 06:15 UTC (12/10) stopped for cleaning focus adjusted again earlier today and looks better now, at -400 Restarted after cleaning at 07:28 UTC 12/10 Dec 10 IFCB not saving files after D20241210T175111. tried re-mapping drive. Activity log showing errors, then connection to instrument randomly stopped. opened IFCB, tried plugging screen directly, did not work, hard power cycle, now working as normal. (re-started at 20:55 UTC 12/10) Evil repeated image is back, in samples D20241211T031826, D20241211T034218 Dec 11 Much windier/big swells/cloudy and rainy today Stopped for cleaning at 06:37 UTC (12/12); re-start at 07:17 UTC 12/12 Dec 12 Repeated image: D20241212T071901, D20241212T074253, D20241212T080644 D20241212T145230 More pennate diatoms today Stopped for a beads run after D20241213T000719. Beads are a new mixture of 50 uL of CountBright Absolute Counting Beads in 50 mL of artificial seawater, which was also filtered through 0.22 um filter w/syringe to remove any remaining salt particles. Beads sample: D20241213T004417 Re-started at 01:15 (12/13) with an extra prime sample at the start. Dec 13 Repeated image: D20241213T093745, D20241213T174053 Not much stuff in the water IFCB connection not working after D20241213T185229. Power cycle at 19:37 UTC 12/13. Did not work; opening IFCB to see if pump is running/things are on. Pump was not on, connected external monitor, restarted, pump on and all seems normal. VNC still not working, unplugged and replugged ethernet cable at laptop and all back to normal. Started again at 22:28 UTC.
